Mutation details: Genomic DNA (including the Mirn150 stem loop with 136 bp of 5' and 130 bp of 3' flanking sequence) preceded by the CAG promoter and a loxP-flanked neo-STOP cassette was targeted to the ROSA26 locus. An frt-flanked IRES-EGFP cassette was positioned between the Mirn150 genomic DNA and the polyadenylation signal. The targeting construct was injected into R19 ES cells that were injected into tetraploid blastocysts. Removal of the floxed neo-STOP cassette via cre-mediated excision (by breeding to Tg(CMV-cre)1Cgn mice) resulted in CAG-driven production of Mirn150 and EGFP in all tissues. This allele shares the same progenitor as allele Gt(ROSA)26Sortm14.1(Mirn150)Rsky and is the progenitor allele for Gt(ROSA)26Sortm2.1(Mirn150)Rsky.
