Mutation detailsThe transgenic construct has the mouse Atoh1 gene under control of the CMV promoter. A lox-stop-lox cassette was inserted between the CMV promoter and the Atoh1 gene in the vector. An IRES-lacZ cassette was ligated downstream of the Atoh1 sequence to monitor Atoh1 expression. (Transgenic mice were generated by DNX Biotherapeutics, Inc (NJ, USA)). Two transgenic lines, M!Z-Tg1 and M1Z-Tg2, were established. No ectopic expression of Atoh1 was detected in the absence of Cre recombination to delete the stop cassette. M1Z transgenics appeared morphologically normal and both lines showed identical phenotypes when crossed to cre-expressing animals. (J:56248)
