Mutation details: The targeting construct consisted, from 5' to 3', of a homology arm with sequence upstream of the Il7r ATG start site, a splice donor site, an intron and a splice acceptor site from the rabbit b-globin gene, the coding sequence of codon-improved cre (icre), a poly-adenylation signal from the rabbit b-globin gene, an FRT-flanked neomycin resistance gene, a 3' homology sequence and a diphtheria toxin subunit A negative selection sequence. Homologous recombination was carried out in E14.1 ES cells. To avoid potential impact of the Neo resistance gene with its enhancer and promoter sequences on iCre expression, the neo cassette was removed from the targeted locus by transient expression of Flp-recombinase, leaving only a single FRT-site in the genome.
