Mutation detailsBAC clone RP23-308B16 was modified by recombineering methods so that a cre-polyA cassette with an FRT-ampicillin-FRT selection cassette replaced the start codon of Mlana (MART-1). Cre expression is controlled by the Mlana (MART-1) regulatory sequences. The targeting construct was electorporated into bacterial cells for homologous recombination and selection, followed by induction of Flp-mediated excision of the ampicillin cassette. The BAC construct was injected into fertilized oocytes and 5 founder lines, 4 of which showed reporter expression in pigment cells when crossed to ROSA26-R mice. Line 5 was propagated for futher analysis. (J:172135)
