Mutation detailsA vector was constructed to insert a strept/FLAG-tandem affinity tag (SF-TAG) followed by a T2A peptide sequence (from Thosea asigna virus) and the codon-improved cre recombinase (icre) sequence into exon 6 of the 1700009P17Rik gene (called Flattop; Fltp). An Frt-flanked PGK-neo sequence followed cre in the construct. The targeting vector was introduced into IDG3.2 ES cells for homologous recombination. Southern blotting showed that recombination occurred on the C57BL/6J allele. After germline transmission was established, mice were crossed to Flpe-expressing line to remove the neo cassette.
