Mutation detailsThe transgenic construct contained the -187 to +247 fragment of the human von Willebrand Factor (VWF) gene followed by the intron-nls cre-polyA sequence. The construct was microinjected into fertilized pronuclei. Four founders were produced, with only 2 (1304 and 1329) producing F1 offspring carrying the transgene. F2 animals were crossed to Z/EG reporter mice and EGFP mRNA and protein expression were analyzed. EGFP mRNA was detected only in the brain of line 1304, but line 1329 expressed in the liver as well and was excluded from further study. EGFP protein was detected in the brain endothelium including some endothelial cells in the choroid plexus overlapping with immunodetection of VWF, but not with GFAP expression, indicating no expression in astrocytes. Line 1304 carries three copies of the transgene. (J:183582)
