Mutation details: The creA fragment of the cre coding sequence (codons 19-59) was fused by flexible F2 linkers to other protein motifs (human H2B and the FKBP dimerizing domain) and a polyadenylation sequence is added. This cassette was cloned into a vector such that it was under transcriptional control of a 12 kb rat Cyp1a1 promoter element. The purified construct was used for pronuclear injection to generate transgenic mice. Beta-naphthoflavone is used to induce the Cyp1a1 promoter so that cre A is produced. These mice are bred with animals expressing the creB fragment of the cre CDS. When dimerization is induced by treatment with AP20187.
