Mutation detailsUsing the piZEG plasmid, a transgene construct comprising the following elements was inserted into the genome: a CAG promoter, a lacZ enzymatic reporter gene cassette with triple poly(A) signals flanked by loxP sites, the coding region from the cDNA of the murine Ptp4a2 gene, an IRES, the EGFP fluorescent reporter gene and a poly(A) signal. The CAG promoter drives the expression of the lacZ genes but not the inserted Ptp4a2 nor the EGFP gene. Only after removasl of the lacZ gene would these two genes be expressed. (J:250112)
