Mutation detailsMice expressing Cre recombinase under the control of the Crhr1 promoter were generated by using a recombinase-mediated cassette exchange (RMCE) strategy. A construct with a docking site contained (5' to 3') an attP site, loxP site, FRT site, splice acceptor, T2A cleavage site, tau/lacZ/flag reporter cassette, FRT site, neo cassette, attP site, PGK promoter and FRT site, was inserted into intron 2. The attB site-flanked Crhr1-Cre recombinase expression unit encompassed, from 5' to 3', Crhr1 intron 2 to exon 3 fused to the Crhr1 cDNA covering exons 4-13, an ires-Cre cassette with a bGH-pA, and a frt site followed by a Pgk1 polyadenylation sequence and hygromycin resistance cassette, both in inverse orientation. phiC31 integrase-mediated cassette exchange resulted in insertion of the Crhr1-Cre expression unit into the right attP site. Resulting ES cells were used to generate chimeric mice, and mouse line. The tau-lacZ reporter and hygromycin selection cassette were removed by breeding to FLPeR mice. (J:264526)
