Mutation detailsThe allele contains four chicken -globin HS4 insulator sequences (two on each ends) flanking the ubiquitous and strong CAG promoter driving LoxP-flanked (floxed) DNA sequence which has the N-PhiM reporter gene encoding a noncytotoxicand non-fluorescent mutated PhiYFP protein with a nuclear localization signal and a transcription stop signal followed by bright membrane-localized fluorescent reporter (MbTomato) and cre gene separated by 2A cleavage site (T2A; mediates ribosomal skipping), then a WPRE sequence, an SV40 polyA signal. A Frt flanked PGK-neo cassette followed those sequences. After recombination/deletion of the floxed cassette by Cre or CreERT2, MbTomato and Cre protein are constitutively active and permanently expressed. (J:278452)
