Mutation detailsA DNA construct with COOH-terminal Cre (cCre)-T2A and Frt-flanked hygro selection cassette were inserted using recombineering into an Nkx2-2 basal exchange vector such that Ccre was fused in-frame with the ATG start codon. The modified Nkx2.2 locus and a hygromycin selection cassette were flanked by Lox66 and Lox2272 elements. This vector was co-injected with a cre-expression vector into ES cells containing an Nkx2-2 (loxed cassette acceptor) allele to generate recombination-mediated cassette exchange targetted allele. Animals were bred to ACTB-Flpe mice to excise the hygromycin marker. (J:311445)
