Mutation detailsA T2A-Cre-Neo sequence was inserted into the 3' of the Tacr1 gene. The targeting vector contained, from 5' to 3', a partial Tacr1 genomic sequence within exon 4, exon 5, Frt3 site, a modified exon 6 with T2A self-cleaving peptide, Cre recombinase, bovine growth hormone polyA sequence, an AttB sequence, a PGK promoter, a prokaryotic gb2 promoter, a neomycin resistance gene, a PGK polyA sequence, a Frt5 site, a mRNA splice acceptor sequence, one domain of the hygromycin resistance gene, a SV40 polyA sequence, and an AttP site, followed by a 3' repetitive Tacr1 arm, lac operon promoter, origin of replication and ampicillin resistance gene. This construct was electroporated into (129S6/SvEvTac x C57BL/6)F1-derived G4 ES cells. Correctly targeted ES cells were injected into recipient blastocysts. Chimeric mice were bred to PhiC31-expressing mice (JAX Stock No. 007743) to remove the AttB/AttP-flanked sequences including the neo. (J:260362)
