Mutation details: An allele with both hypo- and hypermorphic characteristics was generated by the insertion of a neomycin selection cassette in to intron 2. While the targeting vector was designed to incorporate engineered stop codons into exon 2 and thereby completely ablate expression of the four known isoforms, aberrant recombination resulted in the exclusion of the stop codons. Western blot analysis of various tissues showed low levels (less than 10% of wild-type) of the two D1-4 isoforms and increased levels of the two D1,4 isoforms that were double those of wild-type.
