Mutation detailsA loxP site was introduced into the intron upstream of exon 4. A floxed neo cassette and duplicated exons 4 and 5 carrying tyrosine to phenylalanine substitutions at amino acids 182 and 193 (Y182F, Y193F) of the ITAM motif were inserted downstream of the endogenous exon 5. Transient expression of cre recombinase removed the neo cassette leaving behind loxP sites flanking endogenous exons 4 and 5, followed by mutant exons 4 and 5 carrying the amino acid substitutions.
