Mutation details: A targeting vector modified by site-specific mutagenesis to include a nonsense mutation at codon 184 was incorporated at the endogenous locus via homologous recombination. Stopping translation at codon 184 results in the deletion of 45 cytoplasmic amino acids, including those that make up the ITAM. Furthermore, the resultant protein resembles Igh-6 in that it contains only 3 charged cytoplasmic anchor amino acids. An intronic floxed neo cassette was included for selection.
