Mutation details: A neo cassette immediately followed by a 2.3 kb kidney androgen-regulated protein promoter was inserted downstream of the endogenous somatic promoter and upstream of exon 1. The construct was designed such that transcription of the somatic isoform would be controlled by the inserted promoter, which drives expression in proximal tubule epithelium and the uterus. Unexepectedly, expression was undetected in the absence of exogenous androgens. With exogenous androgens, renal expression was less than 2% wild type levels.
