Mutation details: A pgk-neo cassette replaced a 2.2Kb region that contains a putative transcription initiation site and the coding exon for the N-terminal part of the DNA binding domain via homologous recombination. Immunoblot analysis of MEFs confirmed loss of protein. A subsequent publication demonstrated the neo cassette also replaced the first two exons of a second locus located on the opposite strand. This resulted in the loss of the transcription and translational start sites of the second locus. mRNA analysis of numerous tissues from homozygous mice demonstrated transcript levels were 50% to 100% lower than endogenous levels.
