Mutation detailsThe transgene encodes a soluble, dimerizing fusion protein comprising the normal mouse prion protein, without the carboxyl terminal GPI attachment signal, joined to the Fc region of the human immunoglobulin heavy chain gamma 1 constant region (Fcgamma). The construct includes ~6 kb of 5' flanking sequence, exon 1, intron 1 and a fused exon 2/3 from the mouse prion protein gene (Prnp). The Fcgamma coding sequence is modified to eliminate the binding sites for the Fcgamma receptor and complement. A loxP-flanked translation termination (STOP) cassette has been inserted upstream of the open reading frame, blocking expression of the fusion protein until the cassette is excised by Cre recombinase.
