Mutation details: The inducible transgene expresses a dominant-negative inhibitor of CREB, termed A-CREB, by replacing the CREB basic region with an acidic amphipathic protein sequence. The DNA construct was designed to express the following: the first 8 amino acids encode the FLAG epitope tag, the next 13 amino acids are a phi10 protein sequence, the next 31 amino acids are the amphipathic acidic extension, and the final group of amino acids is the mouse CREB leucine zipper, which continues to the natural C terminus of the protein. The mouse CREB B-ZIP domain spans from Leu-274 to Asp-341, the natural C terminus. cDNAs encoding the different B-ZIP leucine zippers and a stop codon were generated by PCR and ligated to sequences encoding the C terminus of the acidic extension.
