Mutation details: Mice bearing this transgene express the highly doxycycline-sensitive rtTA2S-M2 reverse tetracycline transactivator (tet-on) variant under control of the human HNRNPA2B1 promoter. An ~8-kb human genomic DNA fragment was used that spans the CpG island containing the divergently transcribed promoters of the ubiquitously expressed housekeeping genes HNRNPA2B1 and CBX3. Transgenic lines were established from seven founder mice. Line 9 (Tg(Hnrnpa2b1-rtTA2S*M2)9Jstr), exhibiting the highest level of expression detected by northern blot analysis in all tissues examined, was expanded for further study. Transgenic founder 2 had two integration sites, which separated upon breeding to generate lines 2a and 2b; both express the transgene weakly in some tissues. Lines 1, 4, 6 and 7 express low but detectable levels.
