Mutation details: CRISPR/Cas9 technology inserted the full-length human DPP4 cDNA linked to the red fluorescent protein tdTomato by an internal ribosomal entry site into the locus. Expression of the human cDNA is driven by the splice acceptor and A poly(A) sequence and a woodchuck hepatitis virus posttranscriptional regulatory element (WPRE) were added to enhance RNA stability and translation efficiency. Western blot analysis of lung and brain tissue confirmed expression, with highest expression in the lung.
