• Mutation details: Exons 2-4 were replaced by a cassette consisting of a transcriptional repressor sequence (STOP) flanked by loxP sites, cDNA sequence for the first three coding exons (2-4) using native rare codons and including the p.Q61R oncogenic mutation, followed by the 5' 564 bp of intron 4, and an FRT site flanked neomycin resistance gene cassette. The neo cassette was removed via Flp-mediated recombination. Removal of the stop cassette is required to get expression of the mutant Kras at low level due to utilization of the native rare codons.
