Mutation detailsCRISPR/Cas9 technology introduced a serine to methionine substitution at amino acid 82 and a phenylalanine to tyrosine substitution at amino acid 83 (p.S82M/F83Y) into the Ace2em1 zygotes containing the p.H353K change. The Y83 residue in human makes multiple direct contacts with the SARS-CoV-2 RBD. The neighboring position was also humanized due to proximity to F83 and the nonconservative nature of the replacement. These three amino acid substitutions are sufficient to allow infection of mice with multiple strains of SARS-CoV-2. (J:335150)
