Mutation details: Histidine codon 1295 (CAC) in exon 9 was changed to tyrosine (TAC) (ENSMUSP00000096203:p.H1295Y) and aspartic acid codon 1297 (GAC) was changed to alanine (GCC) (ENSMUSP00000096203:p.D1297A) using an sgRNA and an ssODN template with CRISPR/Cas9 technology. These mutations are in the Fe2+ binding pocket of the encoded protein and cause the loss of its catalytic activity.
