Mutation details: Aspartic acid codons 525 (GAC) and 527 (GAT) in exon 14 were changed to alanine (GCC, GCT) (p.D525A, p.D527A) using an sgRNA and an ssODN template with CRISPR/Cas9 technology. The mutations, in the ADD domain of the encoded peptide, reduce the affinity of the peptide to bind to H3K4me0.
