• at E6.5 homozygous embryos appear identical to controls, but at E7.5, about 25% of embryos are morphologically abnormal
• embryos fail to gastrulate
• at E7.5, abnormal embryos fail to differentiate beyond egg cylinder stage; embryos have egg cylinders composed of discrete ectodermal and endodermal layers with no evidence of mesoderm on mixed or 129Sv inbred backgrounds
• no evidence of mesoderm at E7.5
• at E7.5, abnormal embryos show indefinite boundaries between embryonic and extraembryonic regions
