• spleens exhibit a reduction in the number of CD11b+CD1c dendritic cells compared to in wild-type mice
• following E. coli infection, the number of mature dendritic cells in draining lymph nodes is less than in similarly treated wild-type mice
• however, the percentage and numbers of plasmacytoid dendritic cells is normal

immune system
• spleens exhibit a reduction in the number of CD11b+CD1c dendritic cells compared to in wild-type mice
• following E. coli infection, the number of mature dendritic cells in draining lymph nodes is less than in similarly treated wild-type mice
• however, the percentage and numbers of plasmacytoid dendritic cells is normal
• dendritic cells migration is impaired in a dendritic cell intrinsic manner as demonstrated by adoptive transfer experiments
• bone marrow derived dendritic cells lack calcium ion activated nonselective channels unlike wild-type cells
• dendritic cells exposed to E. coli supernatant exhibit increased G protein coupled receptor dependent calcium concentrations due to increased calcium entry compared to similarly treated wild-type cells
• immature and LPS activated dendritic cells are more mobile while E. coli activated dendritic cells are less mobile compared to similarly treated wild-type cells
• however, dendritic cell maturation, differentiation and phagocytosis are normal and treatment of dendritic cells with 2.1M EGTA restores normal calcium levels and migration
• calcium entry after E. coli supernatant stimulation is lower than in wild-type cells due to decreased release from internal stores
