Molecular description: Exon 8 was disrupted by insertion, via homologous recombination, of a TAG3 IRES-nlsCre-neo cassette, which contains stop codons in all reading frames followed by an encephalomyelocarditis virus internal ribosome entry site (IRES), the coding sequence for Cre recombinase flanked by the nuclear localization sequence and polyadenylation signal from SV40, and a neomycin resistance gene driven by the HSV promoter. Cre was shown, both by immunohistochemistry and by X-gal staining of brains of adult F1 progeny from a cross to a beta-galactosidase expressing Cre reporter strain, to be expressed uniformly in nuclei of all cells of the granule cell layer and, additionally, in a few Purkinje cells and in some small cells in the molecular layer.
