Molecular description: The targeting vector consisting of a fusion protein of Cre recombinase plus a modified ligand binding domain of the human progesterone receptor and pgk-neo-polyA cassette was inserted in the translational initiation site of the Grid2 gene in frame. Approximately 900bp nucleotides from the initiation site of the endogenous sequence were deleted. The ligand-binding domain of human progesterone receptor was modified such that it binds and can be activated by the synthetic steroid, RU486, but not by the endogenous ligand, progesterone. Upon induction of inserted cre/PGR expression with RU486, activity was noted in mature Purkinje cells of the cerebellum.
