Molecular description: The targeting vector contains the sequence for cre recombinase and its 3' polyadenylation signal ligated in the opposite orientation to a dual selection cassette (Neo-Tk) containing a neomycin resistance gene and a thymidine kinase gene under control of their respective 5' promoters and polyadenylation. The selection cassette was flanked by Frt sites for deletion in ES cells. Homologous recombination left the Cdkn2a promoter and 5' UTR intact, but replaced most of exon 1Beta with cre cDNA. The linearized vector with a DTA selectable marker at the 5' end was electroporated into ES cells. It was determined that removal of the Neo-TK cassette by transfection of ES cells with FLP recombinase did not result in transmission of the cre knock-in allele, so the Neo-Tk was left intact in the positive chimeras.
