Molecular description: A BAC clone of >160kb from a CJ7/129Sv Mouse BAC library (Invitrogen, formerly Genome Systems Inc.) containing the two exons of Adora2a, flanked by at least 70 kb of Adora2a 5' upstream sequences. This A2AR BAC was modified by homologous recombination in bacteria to replace the sequence of exon 1 (after the ATG transcription start site) with the cre recombinase coding sequence including 3 STOP codons. The purified BAC construct was injected into fertilized B6SJLF1 zygotes. Two founder lines, 2M and 9F, wer established. Mice were crossed to Rosa26-lacZ reporter mice to analyze cre-mediated recombination. Line 2M was used for further studies.
