Molecular description: The creERT2 expression cassette, preceded by a rabbit beta-globin DNA encompassing intron 2 (beta-globin intervening sequences [bGIVS]) and followed by the SV40 polyA signal and a PGK/Tn5-driven neomycin resistance cassette flanked by FRT sites (neo) was inserted by homologous recombination at the transcription initiation site of the Slc1a3 (Glast) gene located in BAC RP23-63O21. The linearized vector was injected into pronuclei of fertilized FVB/N eggs. Line 45-72 showed the highest level of Cre expression, which corresponded to the strong presence of Slc1a3 transcripts. Other regions of the CNS also contained CreERT2 transcripts, but at lower levels. CreERT2 mRNA was also detected in the skin and testis in agreement with elevated Slc1a3 transcript levels in these tissues.
