Molecular description: A -3446 bp fragment of the mouse Gnrh1 promoter was fused upstream of cre recombinase. The proximal region of the Gnrh1 promoter was modified to insert two Tet operators (tetO2). In series, the transgene also contains the CMV promoter regulating the Tet repressor gene (tetR). The rabbit beta-globin intron II is inserted between the promoter and the gene to enhance expression of the tetR gene. In absence of doxycycline, the tet repressor protein binds to the tet operators, blocking transcription. Induction by doxycycline releases the repression, permitting transcription of cre recombinase. Four founders were generated and individually analyzed for function and specificity. A representative line no. 1 was created.
