Molecular description: A targeting vector was designed where the cre recombinase gene was inserted into immediately after the translational initiation site of the Gad1 (GAD67) gene in frame. This vector contained a 3-kb fragment at the 5' side, the cre gene inserted behind the Gad1 translational start, a Pgk-neo-p(A) cassette flanked by two Flp recognition target (FRT) sites, a 7-kb fragment at the 3' side, and an MC1 promoter-driven diphtheria toxin gene for selection. The vector was introduced into C57BL/6 ES cells and ES cells showing homologous recombination were injected into 8-cell stage CD-1 embryos. This allele is a null allele.
