Molecular description: The construct was composed of a 5.8 kb fragment of the nestin promoter with sequences for optimized phiC31 recombinase (phiC31*; coding sequence in this construct lacks the nuclear localization signal) and the tamoxifen-inducible mutated estrogen receptor sequence (ERT2) inserted downstream, followed by the 1.8 kb nestin second intron containing the neural stem/progenitor cell-specific enhancer. The SV40 polyadenylation sequence was also cloned into the vector. The purified construct was injected into fertilized one-cell embryos. 7 founders were established with 2 lines (#2 and 4) chosen for further analysis based on high transgene expression in the ventricular zone of the developing nervous system.
