Molecular description: A construct was generated where four tandem repeats of the A1 enhancer of the aggrecan gene (-9,477 bp to -9,118 bp of Acan) were cloned upstream of the Col2a1 proximal promoter fragment. The cre/ERT2 sequence, followed by an IRES site and the firefly luciferase cDNA, were inserted after these regulatory elements. The construct was validated by transfection into MEFs isolated from Rosa-lacZ reporter mice, and then used to produce transgenic mice. Founders were crossed with reporter-expressing mice and three showed cre-mediated reporter activity. One line was used for further experiments.
