Molecular description: A construct with a 2 kb bovine thryoglobulin (TG) regulatory region, intron II of the rabbit beta globin gene, nucleus-targeted cre recombinase sequence, and the SV40 polyA site was inserted into an Hprt destination vector which has a homology arm matching the 5' upstream region of Hprt and a 3' homology region containing proximal portions of the Hprt gene. Homologous recombination in ES cells with the proximal exon 3 deletion of Hprt resulted in integration of 1 copy of the transgene and restoration of a functional Hprt gene.
