Molecular description: The targeting construct was generated such that exon 2 of the mouse Pdx1 gene in a BAC clone RP23-59N19 was replaced with a modified exon 2 in which a 2A peptide sequence followed by the Dre sequence was inserted immediately before the stop codon of the Pdx1-coding sequence. The FRT-flanked neomycin selection cassette (neo) was placed at the beginning of the 3' untranslated sequence which was removed by crossing chimeric mice with Flpo mice.
