Molecular description: A 208kb BAC containing the mouse Gnrh1 locus (140 kb upstream and 50 kb downstream of the ATG start codon of the Gnrh1 gene) was used to construct a minigene consisting of the fusion protein of modified estrogen receptor alpha ligand-binding protein and a codon-improved Cre recombinase followed by the polyA signal of the human growth hormone gene (hgh pA) and the Frt-flanked neoR (ERt2iCre.FrtNeoRFrt) inserted into exon 2 of the gene downstream of ATG start codon. After removal of the selection marker, the modified BAC DNA was purified and was microinjected into fertilized pronuclei to generate transgenic animals. Eleven transgenic founders were identified. Nine founders transmitted the gene to the offspring. One line (line # 9) was selected, based on its specificity and inducibility of Cre expression, for experiments.
