Mutation details: A neomycin resistance cassette replaced exons 9 to 11, which encode part of zinc finger 1, zinc fingers 2 - 5, and 43 adjacent amino acids. Northern blots of total RNA from E13.5 embryos using a cDNA probe 3' to the zinc fingers resulted in detection of 2 transcripts in wild-type embryos and only one smaller broad band in homozygous mutant embryos. Further analysis by RT-PCR followed by sequencing of the products showed that the mutant allele produced a transcript that spliced around the neomycin insert from exon 8 - 12. The authors state that as a result, the mutant allele could not encode for a protein with DNA-binding activity, but could produce a mutant protein lacking amino acids 269 - 437.
