Cia9 follows an additive mode of inheritance and shows interaction with Cia2.
Mapping and Phenotype information for this QTL, its variants and associated markersJ:69981353 (NOD.Q-H2q x B10.Q-H2q)F2 animals were screened for 155 polymorphic markers at an average spacing of 8.6 cM to identify loci associated with collagen induced arthritis. The NOD.Q-H2q congenic is resistant to collagen induced arthritis compared to the B10.Q-H2q congenic which develops arthritis of intermediate severity at an incidence of 50% following injection with type 2 collagen. (NOD.Q-H2q x B10.Q-H2q)F1 animals are resistant to arthritis. A novel QTL for arthritis severity, Cia9, mapped to mouse Chromosome 1 from 81.6 cM - 106.3 cM with a peak LOD score = 5.6 at the Fcgr2b gene.NOD.Q-H2q-derived alleles at Cia9 contribute to additively inherited disease susceptibility. A locus associated with arthritis severity and C5 serum levels, Cia23, mapped to mouse Chromosome 2 from 11 cM - 37 cM with a peak LOD score = 9.8 at the Hc (C5) gene. B10.Q-H2qderived alleles at Cia23 contribute to additively inherited susceptibility to disease. Cia23 and Cia9 show evidence of interaction: resistance to arthritis depends upon heterozygosity at Cia23 and homozygosity for B10.Q-H2qderived alleles at Cia9. This interaction was confirmed using speed congenic strains carrying a NOD.Q-H2q-derived allele at Cia23 on a B10.Q-H2q background.10.21.2014 Curator's Note: The authors refer to the Chromosome 2 QTL identified here as Cia2. However, because Cia2 was orginally mapped using a DBA/1J x SWR/J cross in J:53356 (1991) we consider the current study a separate mapping experiment and have named this QTL Cia23. |
