Mapping and Phenotype information for this QTL, its variants and associated markersJ:43836A C57BL/6J x (C57BL/6J x C3H/HeJ)F1 backcross population was used in a selective genotyping strategy to identify loci involved in fear-conditioned learning behavior. The C3H/HeJ inbred strain displays poor fear response (time spent in freezing behavior) when introduced to an environment (context) or conditioned stimulus (white noise and light stimulus) associated with electroshock in comparison to C57BL/6J and (C57BL/6J x C3H/HeJ)F1 animals. C3H/HeJ also displays poor fear response when introduced to an altered environment (altered context). C3H/HeJ is homozygous for rd/rd (retinal degeneration) on chromosome 5 but this locus did not show linkage to the 3 behavioral tasks assessed in this study. QTLs associated with contextual learning (freezing to the context) were identified on chromosome 1. Lrnx11 maps to approximately 29 cM between D1Mit123 and D1Mit181 with a maximum LOD score of 5.14. Creb1 was identified as a possible candidate gene for Lrnx11 but there are no known coding region polymorphisms between C57BL/6J and C3H/HeJ. Lrnx12 maps to approximately 65 cM between D1Mit26 and D1Mit15 with a maximum LOD score of 4.76. Suggestive QTLs for contextual learning map to chromosome 3 (LOD = 2.72 at D3Mit106; 55 cM), chromosome 7 (LOD = 1.89 at D7Mit238; 53 cM), chromosome 9 (LOD = 2.70 at D9Mit113; 47 cM), and chromosome 18 (LOD = 1.83 at D18Mit94; 17 cM). A suggestive QTL for freezing to an altered context (pre-conditioned stimulus) mapped to chromosome 8 (LOD = 2.18 at D8Mit4; 14 cM). |
