Epistatic QTL pairs associated with pentobarbital withdrawal were screened in 25 BXD (B = C57BL/6J, D = DBA/2J) recombinant inbred (RI) strains and confirmed in 404 (C57BL/6J x DBA/2J)F2 intercross animals. Parental strain DBA/2J exhibits severe pentobarbital withdrawal whereas parental strain C57BL/6J exhibits mild pentobarbital withdrawal. A QTL on mouse Chromosome 11, Pbw3 (pentobarbital withdrawal 3), was identfied with a modifier locus, Pbwm (pentobarbital withdrawal modifer), on distal mouse Chromosome 1 in both the BXD RI strains and F2 intercross (P = 0.0006). Pbw3 is linked to D11Ncvs69 and D11Mit82 and spans 14 cM - 16 cM on mouse Chromosome 11. Pbwm, the modifying locus on mouse Chromosome 1 is linked to D1Ncvs80 and D1Mit206 and spans 95 cM - 102 cM. Animals homozygous for C57BL/6J-derived alleles at Pbw3 and homozygous for DBA/2J-derived alleles at Pbwm exhibit the highest pentobarbital withdrawal.
