Mapping and Phenotype information for this QTL, its variants and associated markers

Phenotype-driven selection was used to construct congenic strains for the identification of QTLs involved in alcohol preference. Alcohol preferring progeny from a cross between alcohol avoiding donor strain DBA/2IBG and alcohol seeking recipient strain C57BL/6IBG were successively backcrossed to C57BL/6IBG for 7-9 generations.

Provisional QTLs were identified in a genome scan using male congenic animals and 2 of these QTLs, Alcp1 and Alcp25, were confirmed via progeny testing.

Alcp1 on mouse Chromosome 2 has been identified previously. In this study, Alcp1 mapped to 21 cM - 52 cM on mouse Chromosome 2 and gave a peak LOD = 3.7 at approximately 49 cM. The DBA/2IBG-derived alleles appear to have partial dominance at this locus: animals heterozygous at Alcp1 exhibit lower alcohol preference than animals homozygous for C57BL/6IBG-derived alleles.

1.21.2015 Curator Note: Because Alcp1 was originally mapped in J:33277 in 1996 using a (C57BL/6J x DBA/2J)F1 x C57BL/6J backcross, which differs from the cross used here, we consider the current study a separate mapping experiment and have named this QTL Alcp26.

Alcp25 is a novel locus and maps to mouse Chromosome 1 from 6.5 cM-15 cM with peak association at D1Mit65. DBA/2IBG-derived alleles also appear to exert partial dominance at this locus. A possible candidate gene for Alcp25 is Oprk1.
