Mapping and Phenotype information for this QTL, its variants and associated markersJ:52852The genetic basis for differential sensitivity of inbred mice to inflammatory bowel disease induced by dextran sulfate sodium (DSS) was investigated by crossing susceptible C3H/HeJ mice to partially resistant C57BL/6J mice. 86 (C3H/HeJ x C57BL/6J)F2 miceand 90 [(C3H/HeJ x C57BL/6J) x C57BL/6J]N2 backcross mice were analyzed for the presence of QTLs influencing DSS-induced colitis. An additive QTL on mouse Chromosome 2 between D2Mit292 and D2Mit282 (susceptibility marked by the C3H allele at D2Mit94) was significantly linked to several of the colon subphenotypes like severity, hyperplasia, ulceration, and % area involved. The confidence level for linkage of QTL to markers on Chromosome 2 exceeded the threshold for declaring significant linkage (P = 0.0000178). Possible candidate genes in the region include Itga4, Il1a and Il1b. Reduced DSS-induced susceptibility was also observed in congenic stocks in which the highly susceptible NOD/Lt strain carried putative resistance alleles from C57BL/6J on mouse Chromosome 2 confirming the presence of a susceptibility QTL in this region. |
