Candidate Genes
Microarray gene expression analysis was used to identify candidate genes for QTLs associated with collagen-induced arthritis. RNA from inflamed paws of collagen-treated mice were used in the microarray analysis and compared to RNA from normal paws. 223 genes showed a four-fold or greater expression difference between inflamed and normal paws. Nine of these genes are located near previously identified arthritis severity/susceptibility QTLs and may be considered for candidates. Mapping and Phenotype information for this QTL, its variants and associated markersJ:53356The authors performed experiments to localize susceptibility loci for collagen induced arthritis (CIA). F2 crosses between the highly susceptible DBA/1J strain and the highly resistant SWR/J strains were analyzed for the presence of QTLs. A locus, Cia2 on mouse Chromosome 2 had significant evidence of linkage to D2Mit61 (lod score of 6.34). Two-point analysis using BYMARKER showed that the incidence and severity of the CIA phenotype was significantly associated with D2Mit61 (F = 10.5, P<0.0001). Another QTL associated with the age of onset of CIA, Cia4 was also linked to D2Mit61 (lod score of 3.2, F = 15.8 P<0.000001). Candidate genes in this interval include Ptgs1, Dpp4, Ssb and Hc. |
