Mapping and Phenotype information for this QTL, its variants and associated markersJ:7003390 informative markers were screened in 1200 backcross animals derived from parental strains C57BL/FaDk and RIII/FaDk. 300 animals from each of the following 4 backcrosses were inoculated with bovine BSE serum and scored for time of onset of clinical Scrapie symptoms: (C57BL/FaDk x RIII/FaDk)F1 x C57BL/FaDk, (C57BL/FaDk x RIII/FaDk)F1 x RIII/FaDk, (RIII/FaDk x C57BL/FaDk)F1 x C57BL/FaDk, (RIII/FaDk x C57BL/FaDk)F1 x RIII/FaDk. Parental strain C57BL/FaDk displays a mean incubation period of 541 days whereas parental strain RIII/FaDk displays a mean incubation period of 441 days. Four loci affecting the incubation time of prion disease mapped to 4 different mouse chromosomes with a parent of origin effect: progeny of RIII/FaDk mothers display an incubation time 37 days longer than progeny of C57BL/FaDk mothers. Pitm1 (prion incubation time 1) maps to mouse Chromosome 2 at approximately 34 cM (LOD = 5.5 at D2Mit61); Pitm2 maps to mouse Chromosome 4 at approximately 42.5 cM (LOD = 4.5 at D4Mit27); Pitm3 maps tomouse Chromosome 8 at approximately 43 cM (LOD = 5 at D8Mit266); and Pitm4 maps to mouse Chromosome 15 at approximately 49.6 cM (LOD = 3.5 at D15Mit159). C57BL/FaDk-derived alleles contributed to longer incubation time in a dominant fashion at all 4 loci. |
