Injection of mutant ES cells into wild-type (C57BL/6) blastocysts results in chimeric animals with distended abdomens, some of which display premature death at 3 months postpartum. Extensive chimerism (>70% agouti coat color) results in lethality. Chimeras with a 30-70% agouti coat color display: enlarged, polycystic kidneys, renal dysplasia (at 2 weeks), dilated renal tubules (at >2 weeks), and occasional glomerulosclerosis in high % chimeras with severe renal dysplasia. Mutant ES cells fail to contribute to lymphoid and hematopoietic organs (including thymus, spleen, bone marrow and red blood cells) in chimeric mice (J:73254).
