Hpi1 is epistatic to Hpi2. Animals homozygous for C57BL/6J-derived alleles at both Hpi1 and Hpi2 exhibit the highest levels of PMN infiltration.
Mapping and Phenotype information for this QTL, its variants and associated markersJ:69153A genome wide scan was performed to map QTLs involved in hepatic PMN infiltration and necrosis in response to intraperitoneal (i.p.) LPS injection. The analysis of strain distribution patterns in AXB and BXA Recombinant Inbred strains were suggestive in mapping Hpi2 to mouse Chromosome 5. 122 (A/J X C57BL/6J)F2 intercross animals were assayed. Hpi2 on chromosome 5 (spanning 34 cM from D5Mit13 to D5Mit197) was mapped in association with D5Mit233 giving a peak LRS (likelihood ratio statistic) score of 12.7. Reciprocal crosses showed Hpi2 was neither sex-linked nor imprinted. C57BL/6J animals show higher level of hepatic PMN infiltration than A/J, while F1 animals have phenotype similar to C57BL/6J. Hpi2 appears to have no affect in elevating PMN infiltration levels unless another locus, Hpi1 on Chromosome 13, has the B/B genotype. Animals with the highest levels of PMN infiltrations had B/B genotype at both Hpi1 and Hpi2. The authors also suggested the following order of markers on Chromosome 5: D5Mit81 - D5Mit233 - D5Mit391 - D5Mit394. |
