The correlation between gene expression and dietary interaction was examined in a population of (BALB/cStCrlfC3H/Nctr x VY/WffC3Hf/Nctr-A(vy)/a)F1 animals. A/a and A/Avy animals were placed on a 70% (calorie restricted) or 100% (non-restricted) diet andliver mRNA levels were assessed for over 18,000 genes using DNA microarrays. Twenty-eight known genes showing statistically significant differential expression between the 70% and 100% calorie diets and A/a and A/Avy genotypes mapped near known diabesity QTLs. These genes may be considered further for candidate genes.

On mouse Chromosome 6, Smo (7.2 cM) and Flnc (8.5 cM) both show differential expression between restricted and unrestricted diets on the A/a background as well as differential expressionbetween A/a and A/Avy animals on the restricted diet. D6Wsu176e (2 cM) shows differential expression between restricted and unrestricted diets on the A/Avy background. Diabesity QTLs Fglu (16 cM), Mob2 (3.05 cM), and Bwtq3 (4 cM) maps near these candidate genes. Candidate genes for diabesity QTLs Nidd3n (35.5 cM), Obq13 (26.8 cM), Obq14 (43.5 cM), and Bw18 (35 cM) are Mad2l1 (30.3 cM) and Rab11fip5 (35.5 cM, formerly ERATO DOI 32e). Mad2l1 shows differential expression between A/a and A/Avy animalsonboth the restricted and unrestricted diets. Rab11fip5 shows differential expression between A/a and A/Avy animals on the restricted diet.

Mapping and Phenotype information for this QTL, its variants and associated markers

Genome scan was performed on animals from a (C57BL/6J x DBA/2J)F2 intercross to identify QTLs associated with tail length and age-related body weight, and test concordance with QTLs mapped in a previous study. DBA/2J animals are heavier than C57BL/6J animals. Five out of 11 previously mapped QTLs were detected. QTLs for body weight at 3 weeks of age mapped to mouse Chromosome 4 from 40 cM - 60 cM (Bwtq2, LOD = 4.4 at 55 cM), mouse Chromosome 9 from 0 cM - 50 cM (Bwtq4, LOD = 8.3 at 8 cM), and mouse Chromosome 11 from 20 cM - 55 cM (Bwtq5, LOD = 9.5 at 27.8 cM). QTLs for body weight at 10 weeks of age mapped to mouse Chromosome 1 from 40 cM - 70 cM (Bwtq1, LOD = 8.8 at 54.3 cM), mouse Chromosome 6 from 0 cM - 29 cM (Bwtq3, LOD = 4.4 at 4 cM), and mouse Chromosome 15 from 25 cM - 50 cM (Bwtq6, LOD = 7.6 at 41.2 cM). A major QTL for tail length mapped to mouse Chromosome 1 from 40 cM - 70 cM (Tlln, LOD = 33 at 50 cM).
